Article Snippet: 15,000-40,000 cells were grown on 8-well chamber slides for 48 hours prior to fixation with 4% paraformaldehyde (PFA) (Invitrogen) for 15 minutes, permeabilization with 0.5% Triton-X (Sigma) for 10 minutes, blocking with 5% normal goat serum (NGS) (Invitrogen), and incubation with primary antibodies against V5 (Cell Signaling), HA (Cell Signaling), Total FAK (BD Biosciences), pY397 FAK (Invitrogen), and fibrillarin (Invitrogen) overnight at 4°C.
